PTC Reaction of the Month - April 2017

Chiral PTC Addition of Nitromethane to Alkene

By Marc Halpern, the leading expert in industrial phase-transfer catalysis.

This PTC reaction has two very interesting aspects.

First, it is not widely recognized that phase-transfer catalysis enhances the addition of nitromethane to alkenes. This should not be overly surprising since the pKa of nitromethane is 10, so it should be easily deprotonated by common inorganic bases used in PTC systems. In fact, potassium carbonate has a pKb of 10 and was used to induce this reaction in high yield.

Secondly, the inventor wanted chiral addition to the double bond that is not constrained in a ring. This is not easy. The chiral phase-transfer catalyst used, in only 2 mole%, was N-(4,5-dimethoxybenzyl-2-nitrobenzyl)quinidinium bromide, the synthesis of which was noted in this month’s PTC Catalyst of the Month.

Such chiral PTC reactions are usually enhanced by non-polar solvents to push the reactants together in close proximity to the chiral centers of the phase-transfer catalyst. The inventor chose toluene and worked at 0 C to improve enantiomeric excess. The crude product had 76% ee which was increased to 99%+ after recrystallization.
