Chiral PTC C-Alkylation for Cyclopropyl Ring

The chiral C-alkylation described in this patent uses a chiral phase-transfer catalyst has a methyl group on the nitrogen and this is not common for the Maruoka-type azepinium chiral quats. The reaction proceeds well with only 0.5 mole% of the chiral phase-transfer catalyst. This is an unusually low amount relative to other chiral PTC reactions.

As always, chiral PTC reactions are performed in a non-polar solvent to push the reactants and catalyst together into a tighter configuration to enhance chiral recognition.

After the first C-alkylation, the “second alkylating agent” is intramolecular and rearranges. It is interesting that high ee is achieved.

It should be noted that the ester is not hydrolyzed under these PTC conditions. Indeed, phase-transfer catalysis excels in protecting water-sensitive and base-sensitive from aqueous base.

A small amount of the wrong diastereomers is formed (about 3% using the catalyst shown above in Example 12) and is selectively converted into a seven membered diene ring containing a nitrogen, by heating. This facilitates separation from the desired product.
